Please include controls in your experimental design. Signal Transduction Exam 2018 A) The experiment was performed on human embryonic kidney (HEK 293) cell line. The extracts of solubilized proteins were taken under consideration for this experiment. These were then transfected with different vectors: CTL (empty vectors), co-transfected with GFP fused Calmodulin(GFP-CaM) and serotonin receptor either wild type 5-HT2C or 5-HT2CR376/377A which were treated with a vehicle or 5-HT for five minutes, they analyzed the binding reaction by Western Blot and Immunoprecipitation (IP) techniques. Immunoprecipitation was done of these soluble proteins with polyclonal antibody:anti-GFP. Furthermore, these immunoprecipitated proteins were blotted using a monoclonal anti-GFP antibody along with anti-5-HT2C receptor antibody.As a result of this experiment, it was found that the absence of agonist, 5-HT2C receptor did not co-immunoprecipitate with calmodulin in solubilized proteins that were co-transfected with Myc-5-HT2C receptor and GFP-CaM. However, serotonin receptor co-immunoprecipitated with GFP-CaM after treatment with 5-HT for 5 minutes. But the exposure with 5-HT2CR376/377A did not co-immunoprecipitate GFP-CaM with serotonin receptor. It proves that 5-HT2C receptors are linked with CaM depending on the presence of agonist. The experiment performed is to check the agonist dependent precipitation of GFP-CaM and serotonin receptors in the presence of Myc-5HT2C 5-HT exposure. So, the negative control to be designed here for this experiment can be simply of serotonin receptor and CaM. This would result in no immunoprecipitation and prove as a good negative control. On analyzing it through western blotting it could be proved that no immunoprecipitation had occurred in this control. To confirm the results in living cells the same experiment could be used to check the that whether the immunoprecipitation is agonist dependent or not. Take solubilized proteins from living cells and co-transfect them with GFP-CaM and 5-HT2C receptor followed by exposure to 5-HT, then analyze immunoprecipitation via western blotting. The negative control for this experiment will include only CaM and serotonin receptor, and further analyzing it with western blotting, the results of blotting will show no precipitation in negative.2. However, on co-expression of PAG and BRK this band shift was less evident. In figure C: Co-transfection of HEK 293 cells with MYC-PAG and SRC followed by treatment in the absence or presence of SRC kinase inhibitor SU6656 (5 M) for at least 1 hour. PAG was immunoprecipitated by antibody against MYC, and the binding of CSK was compared by immunoblotting. On the other hand, the disturbance of link b/w CSK and PAG was observed on weakening SRC activity by a small molecular inhibitor SU6656, this experiment concluded the importance of SRC kinase activity for functioning of CSK. Also, this study shows that distinct mechanisms have evolved to regulate the activities of two structurally similar and functionally related kinases, BRK and SRC, understanding of the signaling function of protein phosphatases, including definition of their substrate specificities, will allow us to exploit a greater spectrum of the changes in signaling in disease and to generate new and more effective strategies for therapeutic intervention in major human diseasesB) To test the same hypothesis with controls, take breast cancer frozen samples of different patients, divide the sample in two groups one will be the test group other the control group. The control group will be given doses of anti-tumor medication (like tamoxifen for breast cancer). RNA will be extracted from both groups using Trizol and will be followed by purification assay. The breast cancer cell lines will be transfected with empty vectors or pcDNA3-MKP3-V5. Furthermore, the transfectants will be placed in media like minimum essential media MEM with phenol red, followed by SDS PAGE electrophoresis, and transfer to nitrocellulose membrane. After transferring they will be incubated with primary antibody for an hour or more then with the secondary antibody to observe the chemiluminescence with a reagent. The hypothesis being tested here is the enhancement in the lipase activity of phospholipaseC-?1 via phosphorylation of one or both specified tyrosine residues (Y775 and Y783) in the downstream process of signal transduction.To perform the experiment, equal concentrations of purified phospholipase-C-?1 were set on incubation with the active kinase domain of the Fibroblast growth factor receptor FGFR2 and ATP in bovine serum albumin containing buffer, the samples of this reactions were tested for two activities: 1) for lipase activity in the phospholipid vehicles indicated in the figure on left Y axis. This was performed to check the phosphorylation of tyrosine and auto inhibition of PLC-? isozymes, 775/783 of PLC-?1 were substitutes at the place of phenylalanine, they could be used individually or together, but in the experiment tyr783 is used individually. Phospholipase activity of resulting mutant after purification was quantified with active domain of FGFR2K (helps in phosphorylation and activation of phospholipase). Certain known moles of phosphates were added into purified PLC-?1 in wild type under above mentioned conditions and observed that phospholipase activity was enhanced 10 times. The mutation of tyr783 completely nullified the kinase stimulated acceleration of phospholipase activity along with reduction in FGFR2K-promoted phosphorylation of PLC-?1. This experiment proves that, phosphorylation of Tyr783 is vital for relief of auto-inhibition.Studies reveal that Tyr-783 was essential for auto inhibition. As discussed above, permanent phosphorylation of tyr-783 will completely nullify the kinase stimulated and FGFR2K stimulated phosphorylation of PLC-?1. Lipase activity of PLC-?1 will be enhanced across its limits and over-expression of PLC-?1 can induce malignant transformation. The results could be leading to production of carcinoma cells. It has been found in various studies that activity of PLC-?1 is more in cancerous cells as compared to normal cells. Ret signal transduction is increased by translocation of GFR. GFR-mediated Ret activation is essential too for the kidney morphogenesis and for various other functions of abdominal precursors that form abdominal nervous system. Also, GFR has many lipid rafts because its GPI anchorage, but Ret is expelled from lipid rafts. In this paper, the gene replacement for GFR in mice results GDNF resulting in Ret activation but prevented its translocation into lipid rafts. These mice showed renal agenesis, and other disorders including loss of the enteric nervous system, and defects in motor neuron axon path similar to GFR mice that was knocked out, all this provided evidence along with lipid rafts GFR is also needed for neurotrophic factor signaling.Primary considerate neurons secluded from Gfr1and Gfr1TM/TM mice were maintained in vitro for some days. Similar Results were obtained after performing the experiment four times .Moreover, Lipid raft translocation experiments were performed to prove the concept that GDNF/GFR1/Ret complex does not translocate into lipid rafts in Gfr1TM/TM mice. Primary sympathetic neurons from Gfr1/ and Gfr1TM/TM mice were used to extract detergent-resistant membranes. Upon stimulation of Gfr1/ neurons with GDNF, Ret translocated quickly into lipid rafts. This was a contrast to Gfr1TM/TM neurons that an evident reduced movement of Ret into the detergent-resistant was recorded because of GDNF exposure. A small portion of Ret that did translocate into lipid rafts while stimulation may be owing to Ret kinase-dependent translocation of Ret into rafts that occurs with slower movements. There was a significant, 75% reduction in the kinetics of the Ret receptor complex into lipid rafts during GDNF exposure in Gfr1TM/TM neurons according to computation made by these experiments. The negative control design here for confirming the results that Ret doesn't translocate into lipid rafts during GFL activation in Gfr1TM/TM neurons, the primary sympathetic neurons isolated from Gfr1/and Gfr1TM/TM mice will be grown in the same way as test ones, with the only difference that there will be no treatment with GDNF or medium for 15 minutes, and the impact of this will confirm the result to much greater extent upon immunoblotting.

Miller’s portrayal of masculinity Essay

Analyse how the audience might interpret miller’s portrayal of masculinity in act one of â€Å"a view from the bridge†Ã‚  Ã¢â‚¬Å"A View from the Bridge†, is a probing psychological drama. A play script written by Arthur Miller during 1955. It is set in Red Hook Brooklyn New York. It is about an illiterate longshoreman named Eddie Carbone who has relatives come stay with him illegally from Italy who want to persue the America dream. Inspired by a true story of a Brooklyn dockworker who informed on two illegal immigrants. When Miller wrote this script it dealt with water front corruption and graft that questioned the reasonableness of U.S. immigration laws. Arthur Miller was strongly influenced after his father was ruined by depression and after reading Dostoevskys novel â€Å"the brothers karamazor† he decided to become a writer. After reading act one of â€Å"A view from the Bridge† I believe that masculinity is very important to the story because it is closely linked to the American dream and this is the reason why the cousins come to America, to pursue the American dream. Also I believe it is essential to the dramatic tension because if it weren’t for Eddie’s views on masculinity he would not hate Rodolpho, he would accept him and so there would be no tension between them and there would be no story line. This book was written during 1955 and during that time there was a stereotypical view of what masculinity was, it was seen as being strong and having power, authority and responsibility and perhaps demanding respect. It could be that a man was only a man if he had a family and took care of them or if he dressed a certain way. I have added a list on my views of what characteristics should be seen in masculinity: power, strength, authority, responsibility, respect, control, in charge and pride. In â€Å"A view from the Bridge† the protagonist Eddie Carbone shows most of these characteristics at the start but as the story progresses he looses these characteristics and so is seen as less masculine, he looses his pride, authority, leadership he looses the control of his house hold and he looses his respect. In act one of the play masculinity is shown in many different ways, a good example being Eddie carbone. He shows his masculinity in the way he acts. He cares for his family very much, he goes to work everyday or when ever there is work. He says â€Å"he took the food out of his mouth to feed his family† this shows he cares for them and shows his masculinity. Another example of his masculinity is when he drinks a beer or has a cigar this would be seen as a manly or masculine thing to do because women wouldn’t usually drink or smoke. He does this when he comes home after work he sits down and wants a cigar to relax and as a sign of his masculinity and authority, and the respect Catherine has for him she gets it because she has respect for Eddie and he has authority over her  Another reason for Eddie’s masculinity is his work, it could be that working as a longshoreman loading and unloading cargo all day is a different environment from any normal work, it is only men that work at the docks and he is surrounded by men all day so he Is masculine without realising and lifting heavy objects all day could be seen as masculine and only a real man could do this work.

The Five Doctrines of Islamic Faith

Axia college The standard beliefs of Islam are identified as theFive Doctrines of Islamic Faith. They are documented as such, the unquestioned unity of God, the belief in the prophets, the recognition of angels, belief in scripture or koran, and belief in the coming of Final Judgment. Islam is built on the five pillars or acts of worship. These FivePillars describe the expected obligations performed by a Muslim follower. These are described as the basic framework for worship in a Muslim life. The five are as follows declaration of faith, prayer, having concern for the needy, self-purification, and pilgrimage to Mecca. Pillar number one is Al-Shahadaor the declaration of faith. The pillar outlines that all followers must announce their faith to enter into Islam in the presence of witnesses. A kep point of this is knowing that Allah is the only one worthy of your worship. Pillar number two is Salat or prayer, which is required of all Muslims once adulthood is established. Prayer is a daily occurance to be exact five timesper day. Prayer lasts five to ten minutes each time. Pillar number three is Zakat or charity. The islam belief is that all prosperity and wealth is only worthy of God, and God’s desire is for Muslims to express generocity in life. Zakat requires the giving of a piece of a persons financial wealth to charity at the conclusion of the year. Pillar number four is Sawm or fasting. Every Muslim is required to fast for Ramadan which falls at month number nine of the muslim calendar. The muslim reasoning for fasting is to cleanse you spiritually and physically as well as focus your attention on spiritual aspects rather than on physical ones. Fasting for Muslims includes refraining from eating or drinking anything as well as having sexual intercourse from dawn to dusk. The last Pillar is Hajj or the pilgrimage to Mecca. Every Muslim outside of those who are physically or financially unable, must makethe pilgrimage to Mecca at least once during the course of life. My thought is that the declaration of faith would be the easiest because its a one time pillar that takes a short time. The most challenging would be the fast from dusk to dawn because I would find it difficult to focus all day with no food for 12 hours or so.

We have no way of knowing what causes them. For Hume, if an idea has no corresponding impression, then it is meaningless, that is, it does not exist. Kant’s ethics distinguishes between â€Å"acting in accordance with duty† and â€Å"acting for the sake of duty.† The latter are actions that have moral worth, while the former has no moral worth. Moreover, Kant explains another way of determining whether an action is morally good or not. This is presented in The Categorical Imperative. This principle states that before I will a certain mode of conduct, I should see to it that my subjective course of action could be universalized as a moral legislation. Ageism in the Workplace Essay Ageism can happen to anyone, young or old. Many times when discussing Ageism or age discrimination, most people consider older generations as those affected, but the reality is that younger generations can be affected by Ageism as well. Ageism is growing in America today, and there are many significant issues that are being associated with this form of discrimination. The purpose of this paper is to define Ageism and who is being affected by it. Those affected are experiencing a wide array of issues that can ultimately have serious and sometimes fatal consequences. Ageism is not something that cannot be cured, and there are definitely ways to overcome it. Ways to combat this problem, include education and mentoring. This paper will outline many different opportunities within these two areas and ultimately conclude with recommendations for organizational management to combat the growing issue of Ageism. Ageism Definition What is Ageism? In simple terms, ageism is defined as a form of discrimination targeting older adults, generally 55 years of age and older. Dr. Linda Woolf uses a definition by A. J.  Traxler to define Ageism as any attitude, action, or institutional structure which subordinates a person or group because of age or any assignment of roles in society purely on the basis of age. Ageism is different from other forms of discrimination in that an individual’s age is static unlike their race or gender (Woolf, 1998). What Dr. Woolf means by this is that a person’s age is constantly changing, and no one is exempt from experiencing ageism in some form. All of us at some point will be affected by Ageism, whether we stereotype others or we are stereotyped based simply on how old we are at that point in time. Ageism is slowly becoming a major issue within the workforce of America. Persons Affected Baby Boomers. There are some discrepancies in which period of time the Baby-Boomer generations actually begin and end, but for my research, the period is established as 1946 to 1964 (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). Baby-Boomers were born in a five to six year period immediately following World War II. The current age range for Baby-Boomers would be around 63 to 68 years of age. This generation is obviously within the prime age range to be affected by Ageism. The consulting firm, Age Lessons, lists 3 R’s as areas of concern for Baby-Boomers: redundancy, relevance, and resentment from younger co-workers (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). Boomers definitely have reason to be concerned about redundancy. In this day and age with so many layoffs occurring, many companies are looking at older employees, especially those within retirement age, as individuals to cut. The Boomer generation, having been around a while, most times is more expensive to retain than younger employees. Many older employees struggle to remain productive and relevant within the organization. They fear being left behind unless their employer offers them training opportunities (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). The final R, and maybe the harshest, is resentment from younger employees looking to grow and advance within an organization, but feel they are being held back by the Boomer generation. I have seen this type of resentment first hand, and it can and does often lead to an unstable work environment. Generation X. Who makes up Generation X? Well there are many different ranges of time given to define this group of individuals, but a good estimate is 1965 to 1980 (Jochim, 1997). The age range of this group is now around 33-48 years of age. That doesn’t seem very old, and one would wonder how Ageism could be affecting this group. They would seem to be in their prime working years. The reality is that as of 2003, more and more young workers feel they are being discriminated against because of their age (Armour, 2003). Revealing statistics support this belief among young workers. In an article for USA Today, Jennifer Armour reveals that in 2003 the unemployment rate for workers 25 to 34 was at 6. 9 percent, well above the 3. 9 percent for employees 55 and older. A more recent poll by Gallup, as of April 2012, puts the underemployment rate for adults 30 to 49 at 14 percent, still ahead of the 13. 6 percent for adults 50 to 64. Compare both of these with a 12. 7 percent rate for adults 65 and older, and you can see that ageism can affect all ages. This is a startling increase since 2003 and speaks to our current economy and the mindset of employers when determining who to retain and who to let go. With so many young adults underemployed or unemployed, they are unable to attain the experience and skills necessary to help them grow and attain better jobs in the future (Jacobe, 2012). These issues combined could lead to significant repercussions for the future of American business. Issues of Ageism Health There are many issues that lead to Ageism in the workplace. One issue is the health of the employee. Generally, organizations worry about the health of their older employees and how it may relate to work attendance. One stereotype is that older workers seem to be more fragile and are not able to physically perform as well as their younger counterparts (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). Issues of Ageism surrounding employee health are not only limited to physical requirements, but include mental aptitude as well. In fact, a 2001 survey conducted at Duke University found that nearly 80 percent of respondents age 60 and older experienced ageism such as people assuming they had memory impairments because of their age (Dittmann, 2003). Costs Many costs are associated with Ageism, but they are not all economic in nature. Costs to the elderly, costs to the younger generation, as well as social costs are a few that contribute to the problem. The discrimination of Ageism can affect individuals in many ways such as being denied employment or being passed over for promotion, just to mention a couple. One significant, yet subtle, blow can occur to the individual’s self-esteem (Palmore, 2005). The feeling of no longer being needed or wanted within an organization can have significant consequences to an elderly individual. It can lead to depression and a rapid deterioration of mental and physical skills. At the other end of the spectrum, is the much younger generation of workers. The costs of ageism can affect them as well. The costs to them are related to the topic of economic costs. According to Mr. Palmore, special programs that benefit only older persons, such as Medicare and Medicaid, are funded by the tax dollars of the younger employees within the workforce; the amount was greater than $300 billion annually in 1999. The final cost is the social cost to not only the elders, but the younger generation as well. The younger generation loses a significant wealth of knowledge to pull from while the elderly lose an opportunity for social interaction. Technology As time goes by, technology continues to progress at a rapid pace, and it is often difficult for individuals, as well as organizations, to keep up with the ever changing products. Many managers feel that older workers do not like change and lack the desire to learn new technology or simply have no valuable technical skills to offer (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). In my own personal experience in the technology industry, I have supervised older employees who had the minimal skills to succeed but did not want to put forth the effort to learn new skills in order to advance. Likewise, I have had supervisors who are â€Å"old school† and had no interests in keeping up with the technological advances. This led to younger employees actually having a greater knowledge base than their manager. Education Education of older employees is also a concern for managers in today’s constantly changing environment. The changes of the work environment have brought new knowledge, skill, and ability requirements for all workers (Lee, Czaja Sharit, 2009). The question that managers have to answer is, do older employees have the necessary cognitive abilities to learn and perform these new requirements. As I mentioned earlier, some older adults do not have the desire to learn new skills at such a late stage of their career, and this is an issue many managers are faced with. Attitude can play a large part in a manager’s decision to select older adults for the necessary training to keep those individuals gainfully employed. A negative attitude by older employees can lead to negative stereotypes and concerns about the trainability of such individuals (Lee, Czaja Sharit, 2009). Older adults, who display a desire and enthusiasm about learning new skills and abilities, have a greater opportunity to overcome the negative stereotypes and contribute significantly to any organization. Risks While efforts are being made to eliminate all forms of discrimination, it continues to occur today to many groups including older adults. Wendy Taormina-Weiss states, â€Å"Despite efforts to provide a level of protection that might be based upon compassion, ageism continues to lead to disempowerment of seniors in this nation. † The question becomes, what are the risks we are creating for seniors by discriminating against them because of their age? Poor health, reduction or loss of financial security, and social isolation are three major risks created by ageist beliefs (Taormina-Weiss, 2012). Many seniors have lived a long and productive life, and the need to make a contribution is important to them. By looking at them with these negative stereotypes and discriminating against them simply because of their age, the risk factors above can have severe consequences. Taormina-Weiss goes on to say that these risk factors can contribute to a lower quality of life, lower self-esteem, and ultimately a shorter life span. Combating Ageism Education Outsourced. The Age Discrimination in Employment Act (ADEA) prohibits age discrimination in the workplace for employees or potential employees 40 years of age or older. (Farrell, 2011). Assistance, such as Professional Employer Organizations (PEO), is available to employers to assist with understanding and compliance of the law. According to PEO Spectrum, one benefit of a PEO is that they co-employ with a client accepting half the risk of managing an organizations employees. Just as important, is the fact that PEOs provide training to supervisors and managers to not only improve employee communication and relations, but also to keep leaders up to date on applicable laws pertaining to age discrimination (Farrell, 2011). Other areas that PEOs offer training in is proper interviewing techniques, managing through a layoff, discrimination, and harassment (Farrell, 2011). Organizations should have liability protection insurance for a worst-case scenario such as a wrongful termination. The issue here is that liability insurance can be expensive. According to Farrell, most PEOs offer their clients coverage, but the cost can be as much as $5000 a year for a company with only 10 to 20 employees. However, with the proper training and education, such as that provided by PEOs, organizations can significantly reduce the risk of facing such a lawsuit. In-house. In a study conducted by several faculty members and students at Nova Southeastern University on the subject of Ageism, the authors recommended the TEAM approach in order to build diverse high-performing teams. The TEAM approach stands for team composition, education and training, awareness, accountability, and accommodation, and mentoring (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). Team composition is the first and most important step. When building the team, you should ensure to include employees across all age groups. Education and training are also vital to your employees. Diversity training has become very important along with opportunities for older adults to receive training to keep technical skills current. Research shows that the 2008 U. S. Presidential election was notable for many things, mainly the difference in how each candidate reached their constituents; one used email and text messaging, while the other did not (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). Organizations need to be aware of discrimination laws and ensure that all employees are aware of the requirements to be in compliance with said laws. The Human Resource department of an organization must be responsible for developing strategies to ensure that all employees, especially the Baby-Boomer generation, are given the same opportunities for performance and promotion (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). The final peace of the TEAM concept is mentoring. Mentoring can be a two-way street in any organization. Not only do the older adults have an opportunity to mentor and train the next generation of employees, but the younger employees have an opportunity to mentor and train older adults to keep them current on new technologies and techniques. Mentoring Management. There are many opportunities for mentoring members of an organization’s management team. Many professional services are offered as well as opportunities from within an organization to use firsthand experience and knowledge. As discussed in the previous section, mentoring can and should occur in both directions. Knowledge should be gained from those adult co-workers who have been with an organization or industry for multiple years and have unique ideas and insights. Conversely, the younger generation of leader may bring a fresh set of ideas and beliefs that could make the organization more efficient. Two key areas of opportunity for mentoring managers is the use of Senior Mentors and Peer to Peer mentoring. Senior Mentors. Our own U. S. military is a good example of using Senior Mentors to assist individuals in upper leadership positions. These mentors help conduct exercises and offer advice to their still-active colleagues (Vanden Brook, Dilanian Locker, 2009). The majority of these retired leaders has experienced what our new leaders are about to experience and can offer valuable advice on how to handle certain situations and what steps to take in certain scenarios. While I could not find statistical data to show that the program has been effective in the military arena, I have no doubt that having the insight of someone who has held the same position previously is nothing short of valuable. Many of the same principles applied in military circles can be applied in the business sector as well. Retired military leaders know all too well the issues of discrimination, no matter what form, and can help guide the next generation of leader in avoiding these pitfalls. In the military, we sometimes find Ageism pointed in the direction of the younger generation because of a lack of experience. This scenario is where Senior Mentors can advise leaders to listen to their younger leaders and avoid the appearance of prejudice against them simply because of their age. Peers. According to Judith Germain, peer to peer mentoring occurs when the mentor is not in a position of authority over the mentee. Peer to peer mentoring can be very valuable in a group setting because mentors are able to pass on knowledge based on life experience and professional experience gained in the business arena (Germain, 2011). Too often we think of a peer as someone similar in age, but in the business world it relates directly to job title. Putting peers together from different age groups to exchange information and ideas goes a long way in helping them understand each other and eliminating any stereotypes about each other. Germain goes on to say that in this group setting with such a diverse group of leaders, the learning curve for the business owner is greatly reduced. In the long run, peer to peer mentoring offers a relaxed environment among managers of all experience levels to pass on valuable life experience and information. Employees. Mentors use their leadership to advise and nurture their employees either voluntarily or involuntarily (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). As discussed earlier, managers and leaders need mentoring before they can mentor their employees. The information held by the leaders of an organization must be transferred to the next generation in order to assure continuity of key organizational functions (Gibson, Jones, Cella, Clark, Epstein Haselberger, 2010). When related to Ageism, managers must mentor their younger employees/supervisors concerning discrimination laws so that any issues with age discrimination may be avoided. Communication should be imperative in the mentoring process. With the proper mentoring program in place, valuable knowledge can be transferred, new and valuable friendships can be formed, and unnecessary litigation can be avoided. Conclusion Ageism is a rapidly growing form of discrimination. As we have learned, many Baby-Boomers today are experiencing or already experiencing this form of discrimination. Ageism is not limited to the Baby-Boomer generation, however, as many younger generations are experiencing this prejudice as well. Many factors have been discussed here including health related issues, social issues, and financial issues that arise out of this act of prejudice. These factors have a negative impact on employees of all ages and can have a significant negative impact on an organization if steps are not taken to correct any form of age discrimination. The good news for any organization is that there are ways to combat this growing problem. Organizational management should conduct an audit of policies and procedures to determine if they have allowed for any opportunities for this form of discrimination to occur. If so, they must take the necessary steps to correct the issue. Management should take advantage of every opportunity to educate their staff from top to bottom on applicable state and federal laws as well as internal policies and procedures related to any form of discrimination. Finally, it is recommended that management create and support a company mentoring plan. This plan should use outside resources, such as Senior Mentors, as well as current employees of the organization. With the proper policies, procedures, education, and mentoring programs in place, an organization can avoid any potential pitfalls of Ageism or other forms of discrimination.

The Cuban Identity: truly their own? The United States became involved with Cuba's independence struggle with Spain because they had a keen interest in extending their power into offshore colonies. Although the war had started three years prior to the United States' declaration of war on Spain, they began to exert their influence and power over the colony by infusing a government structured by appointees of President William McKinley. These authorities created the interim Cuban government as a subset of the United States and the politics and social structures of which resembled the United States rather than focusing on a Cuban specific structure. They created a government based on democracy which then drew from the social structure of Cubans of different color voting for positions within this government. Their structure of the Cuban system was strikingly similar to chang es that were being undertaken in Louisiana at the time and Cuba was a secondary location to exert similar practices. McKinley appointed General John Brooke to be the first military governor in 1899 and Brooke had been known for protecting the liberties of black voters in the United States (Scott 155). Women have played a large role in this changing of principles and ideals, creating rights that they deserved but did not always have. One such right is the right to present and obtain a good education through the home and the public system. During the early 20th century there has been a conscious move by society to allow for a greater opportunity for women to become better educated and to educate others. This form of a women's movement began in the home and pushed its way through the public system. We have the men and women in southern Arizona of the early 20th century to thank for the opportunities in education presented to both genders, but more specifically women, in the present day. It is important to say why education is such a necessity. History has shown that the more educated person is the one who survives the longest, is more successful in business, and overall enjoys life more than a person who is non-educated. Women have not always been offered a good education because education brings on new ways of thinking, and in a male dominated society (such as the U.S. was in the early 20th century) this can create havoc in the home and in society. Just as slave owners in the south did not want their slaves to become educated for fear of rebellion, this male dominated society did not want women to become educated because they would want more and more of what the men had. Kean University

I never thought this day would come as fast as it did. I feel like this year is just going to fly by and the next thing I know I’ll be packing to go to college, but let me not get ahead of myself. I remember always being asked, â€Å"What do you want to be when you grow up? † The funny thing is, I’ve always known. I’ve always wanted to be a teacher. I can look back to my younger years and remember playing teacher, writing out worksheets, teaching classes with my cousins. It’s something I always enjoyed. I am apart of the Coop Education program at my high school, I am now an intern at a preschool. It is just one step closer to what I want to do in my future. I now have hands on experience with young children, being able to interact and add input with any ideas I may have. As you can see, I am a determined young lady; if I want something I am going to try my hardest to be the best I can be in the field I am interested in. Kean University has so much to offer me, especially because I know what I want to do with my life. Kean is a teaching University and elementary education is one of the most popular fields at Kean. Why else would I look any further? I have a plan, people with plans become successful. I am someone who can be a team player when needed be. I was on my schools softball team my sophomore year in high school. I believe that playing a sport, taught me sportsmanship, self-confidence, and being able to work as a unit. You are able to bond with you teammates through the rough long practices and demanding games. I enjoy being involved in school activities. I was apart of Spanish Club and was the secretary of this club in 2007. We had fund raisers, put on a show for the International Festival two years in a row, held at Bloomfield High School, and also provided semi formals ,many accomplishments were made being apart of the Spanish Club. I was also joined Stage Crew my junior year, it gave me insight as to how plays and shows are put together. It’s not an easy job; it is time consuming but a learning experience. Next time I watch a play, I know how much hard work everyone backstage put in, being a part of stage setup and lighting was something new I tried out and did enjoy. I am a determined young woman ready to attain new information in a new ambiance with different people from different walks of life. I am ready to leave my nutshell and burst into a new atmosphere I can say Kean University is where I would like to spend the next four years of my life growing. The campus is big itself, and is unlike any environment I’ve put myself in. I would like to dorm. It’s a time in my life were I can be independent. A year away from my parents and around people I am not familiar with . Life is about experiencing new things. I want to advance, study and experience this all at Kean University

To Free the Slaves When the nation was founded, the southern stated favored slavery and the northern states did not. Had the presidents that came before Abraham Lincoln stood up and made the much needed change, then it would not have fallen on Lincoln’s shoulders to stand up and free the slaves. Because this duty was laid upon him, Lincoln took many steps to abolish slavery. Among the many things that he did was to stand up during the Civil War, enact the emancipation proclamation, and pass the 13th amendment. Had he not taken these steps to abolish slavery, history would have been completely different, and the road to civil rights would have been delayed longer than the hundred years that it took to begin achieving equality. On November 6, 1860 Abraham Lincoln was elected the President of the United States; this event caused outrage among the southern states. Because Lincoln had run on anti-slavery, the southerners believed that they no longer had a place in the Union. By February 1st, 1861 seven states had seceded from the Union. The seceded states collaborated to create the Confederate States of America and elected Jefferson Davis as their president. Lincoln decided to fight because he felt that it was his duty as the president to preserve the union. During his first inaugural address he appealed to the rebellious states to rejoin the union. (Civil War) The Civil War began on April 12th, 1861 in South Carolina at Fort Sumter. The Civil War was not entirely caused by the election of Lincoln. However, it was the primary reason the war broke out when it did. It also in my opinion set the stage for abolishing slavery. At this time Lincoln upheld that blacks could not join the army, however they were allowed to fight for the navy. Many blacks rushed in to fight because; although Lincoln stated that the war was not about slavery, but instead about the union. The African Americans knew this war was about slavery (Handlin). Slaves were escaping in masses, and because there was no consistent law regarding fugitives, commanders could make their own decisions on what to do with them. Some were put to work for union forces, some returned to their owners. Then on August 6th, 1861, the fugitive slaves were considered to be â€Å"contraband of war,† and it was decided that if they fought, and they were found to be contraband of war then they were declared free. As they continued to move south many slaves flooded the north. As a result both white and black northern citizens pulled together, providing aid, and organizing relief. And throughout the war they also organized schools to teach the African Americans to read and write (McPherson). Then in 1862 Lincoln began considering emancipation as a stepping stone toward winning the war. In the south they began to force blacks to build forts, work as blacksmiths, nurses, boatmen, laundresses, and work in hospitals, armories and factories. However, in the north the African Americans who wanted to defeat the slaveholders the most were being turned away from the military, because in the beginning of the war Lincoln did not want the south to believe that the war was about slavery not the not the Union. At this time, if Lincoln declared that the war was about slavery, European public opinion would back the north. On September 22nd, 1862, Lincoln issued the preliminary Emancipation Proclamation. The proclamation warned that if the Confederate states did not surrender by January 1st, 1863, their slaves would be freed. Many believed that this was the beginning of the end of slavery. But, they also feared that Lincoln would give into pressure and fail to keep his promise. Then on January 1st, 1863, he issued the final Emancipation Proclamation, freeing all slaves in states that were not part of the union. At this point, the purpose of the war changed; they were not only fighting to protect the union, but also fighting to end slavery. After the final draft was issued, the federal army began to officially accept black soldiers into ranks (Goodwin). A new chapter of history began, when the Thirteenth Amendment passed in January 1865. This Amendment abolished slavery in the United States, and provided that â€Å"Neither slavery nor involuntary servitude, except as a punishment for crime whereof the party shall have been duly convicted, shall exist within the United States, or any place subject to their jurisdiction. (McPherson) The Amendment was passed at the end of the war, before Confederate States had rejoined the Union, making it easy to pass. Finally, on April 18, 1865, the Civil War ended with the surrender of the Confederate army. The southern landscape was destroyed, and four million slaves were freed. Former slaveholders faced emancipation with rage, concern, and despair. The sl aves were freed and began to attempt to rebuild their lives. The country then began reconstruction, which held many promises and tragic disappointments for the people of the United States. As part of the reconstruction there were two new amendments added to the constitution. The Fourteenth Amendment, which granted to all people born or naturalized in the United States. And, the Fifteenth Amendment, which said that no American can be denied the right to vote on the basis of race (McPherson). The Civil War paved the way for President Lincoln to abolish slavery in the United States. First he took a stand; he enacted the Emancipation Proclamation, and finally he was able to pass the Thirteenth Amendment. Had Lincoln not taken these steps to abolish slavery, History would have been completely different, and the road to civil rights and equality would have been delayed much longer. We will never know who would have finally stood up if he hadn’t, nor how long it would have taken. However, we do know it would have been different. Would we have had Martin Luther King? Poetry has been around for ages and enjoyed from children to adults alike. Poetry is not just words on paper that imparts data; it is much more than that. Poetry is an art form that in order to be fully understood, one has to be able to analyze read between the lines. Analyzing poetry can be a daunting task. One may have to read the poem several times with a dictionary handy, just to get an idea what the poem is about. Some poems are made of elaborate statements while only using as few words as possible. The beauty of poetry is being able to visualize what the writer is trying to communicate to the reader and putting it into their own words and scenario. â€Å"My Papa’s Waltz,† by Theodore Roethke can be interpreted in many ways. At first, it can be construed as a child writing about his alcoholic father and the pain that was inflicted on the small boy by the father’s actions. After rereading the poem, the reader can actually see that the father is frolicking about with his son, he may have had a little too much to drink, but that does not depict alcoholism nor abuse. Roethke was shattered when his father died suddenly of a heart attack, when the young man was only fifteen years old. This poem shows him reminiscing about the good times he had with father. â€Å"White Lies,† by Natasha Tretheway is a touching poem about a young girl dealing with her skin color and trying to find acceptance in a judgmental world. A reader can feel the little girl’s pain, loneliness, shame, and fear as the poem is read. Tretheway mentions colors several times in the poem, which alerts the reader on the importance of color throughout. â€Å"I could easily tell the white folk/that we lived uptown, / not in that pink and green/ shanty-fied shotgun section/ along the tracks.† (7-11) for a little girl to feel so ashamed of whom she is and where she came from is heartbreaking and Tretheway put it into perfect detail. â€Å"My Childhood-Home I See Again,† by Abraham Lincoln is a two-part poem that  deals with the issue of memory and having a sense of ourselves in relation to the past. Lincoln used descriptive words to pull the reader into his life and see what it was like for him to grow up. Lincoln wrote the poem after visiting his home of Pigeon Creek, Indiana in 1844. Twelve years after the poem was written, Abraham Lincoln was elected the sixteenth president of the United Sates. Poetry can tell a story about many things. The poet has a story to tell. How the reader interrupts that story is up to them. The reader may not get out of the poem what the poet put into it. I was inspired by this quote that speaks of success, which I will describe as a golden star that everyone wants to reach. This quote is similar to most of the leadership quotes because it speaks of ways to reach success, but, this quote is special in a way that it could be related to the self-fulfilling prophecy. Simply put, the main road to success is the â€Å"will to succeed. † I’m sure each of us has experienced actually failing a task whenever our mind is embedded with the phrase: â€Å"I can’t do this! † If you believe that you will be successful in any endeavor you want to take, then, you have taken the first step to becoming a person who is the â€Å"king or queen of success. † I myself have noticed that whenever I put into mind that I will â€Å"win,† the outcome turns out to be truly positive. This essay can help readers realize that their goals can be achieved by first believing in themselves. If a person always expects that he/she will succeed in something specific, then his/her expectations will most probably turn into a reality as what the so-called â€Å"self-fulfilling prophecy† suggests. He grew up in Hodgenville, Kentucky, and lived out in the wilderness. He was brought up doing chores, and had a special talent of using an axe at a young age. Upon attending an ABC school, Abraham learned how to read, write, and solve arithmetic problems. He and his family constantly had problems with the milk sick disease, which was the cause of the death of Abraham’s mother. One of Abraham’s first jobs as a young adult was to build a flatboat and take it down the Mississippi River loaded with cargo. From then on, his occupations seemed to be constantly changing. Some of the other jobs he became involved in were a clerk in a general store in New Salem, served three months as a private, opening a general store in New Salem once again which failed after a couple months, state legislator of Illinois, and a series of other more political jobs. Abraham Lincoln married twice; first to Ann Rutledge and later as a result of her death e married Mary Todd Lincoln. They had four children, and only one reached adulthood. Lincoln’s first real successful career was as a frontier lawyer, in which he earned about $1200 to $1500 a year. Eventually seeking higher political positions, he became a powerful young congressman. At this time in his life, her first began to have the opportunity to voice his opinions about slavery. He was very much against it, and supported Wilmot Proviso, which proposed that slavery be abolished from Mexico, and he formed other antislavery programs in Washington. Upon returning to his former job as a layer upon the end of his political term, he was able to become a master at the occupation. Around 1854, Abraham began to research the Kansas-Nebraska Act, for he was held an opposing viewpoint of what is stated. He believed the Act did not address how important it was that slavery be abolished, for it allowed people in these two territories to decide for themselves whether or not to allow slavery within their borders. Right after this, Lincoln began involving himself in more debates about slavery, while at the same time trying to get into a political position. He was finally granted his wish to become president on November 6, 1860 defeating Douglas, John Bell, and John C. Breckinridge. Re-elected in for the next four years after this term had ended, he was all the while serving during the Civil War. His view on the war was that he hoped to create a plan to join the nation together, and give the South more right to the slaves. Eventually the Civil War’s end was exactly how he had wanted to terminate, with General Lee surrendering to General Grant in Virginia. The death of President Lincoln was tragic, and was soon following this victorious end of the war. Upon a third re-election as U.S. President, he angered many people who did not like the way he served their country. John Wilkes Booth was one of such people, and he expressed his anger during a performance of â€Å"Our American Cousin† at Ford’s Theatre in Washington, when he assassinated President Abraham Lincoln. Throughout Abraham Lincoln’s life, he was succ essful in politics aided in the progress of our nation. Abraham played many extremely important roles in this particular time period of history, while his most prominent position was undoubtedly President of the United States. During his role in presidency, many Southern states were against his election, including Mississippi, Florida, Alabama, Georgia, Louisiana, and Texas. Later four more states were angered and all ten succeeded and left the Union around 1860. With no decision but war, the northern and the southern states were allowed to fight over issues dealing with slavery, rights, and preserving the Union. On January 1st, 1863, President Lincoln issued the Emancipation Proclamation, which entitled that all citizens who were slaves up to now are free. The importance of this declaration was not that it freed many blacks in the South, but that it showed Americans that the present war was being fought to end slavery. Besides this major point, the Emancipation Proclamation allowed black men to feel accepted and equal during the war, for they were allowed to hold better positions and could enroll in the Union Army and Navy. The Gettysburg Address was later given by President Lincoln as well, which where he defined the Civil War as a rededication to the ideals of the Declaration of Independence. This was one of his most well known speeches, for the site at which he announced it was a memorial for the death of soldiers. President Lincoln was indeed a significant person to the history of the United States and its overall development. He is probably the President with the most information documented about his life, and he is remembered today as a man who gave touching speeches and who fought endlessly for what he believed in. He always put forth his efforts into ending wrongs and never giving into what is morally wrong. Because he was committed to preserving the Union no matter what the consequences were to himself, it was saved, and because of his patience, timing, calculations, and keenness at solving problems he was able to kill what was left of slavery. Through transforming the president’s role as commander-in-chief and as chief executive of his nation into a more powerful position, presidents from his term in office on were granted supremacy over both the Congress and the courts. His perseverance as a person brought him to have such an influential remembrance of him today, as he started out as a poor boy brought up in the wilderness in a log cabin to obtaining the most revered position of the country in the White House. Although he ended a tragic death, America will never forget his efforts at making America a better place, and will always look towards him as the father of our country.
